Pfizer Paper – Production of rAAV by plasmid transfection induces Antiviral inflammatory responses in suspension HEK293 cells

Innate cellular defenses are a persistent barrier to viral vector manufacturing. The article shows that innate cellular defenses, triggered by plasmid transfection during rAAV production in HEK293 cells, cause antiviral and inflammatory responses. This innate response, including activation of interferon-stimulated genes and pathogen recognition receptors, limits vector yield and highlights opportunities for improving production efficiency by managing these cellular pathways
